How do I prune to promote growth to fill in a hole in my privet hedge? Do I clip around the edges of the gap to encourage new growth? There is a low spot right in the middle of the 25' long hedge... is it necessary to cut the rest of the hedges down to the same height?; I would rather not lose all that growth.

The adjoining plants should fill in over time either way, but clipping along the edges of the gap will encourage additional branching there. Note that you will need to stop pruning by early fall so the growth has time to harden off before winter.
