I am expanding my garden. I am tilling over an existing lawn. I want to know if I should use mushroom manure or something else to get the soil ready for planting. Thank You.

Mushroom compost and other organic materials (such as old rotted leaves, rotted stable manure and bedding, compost, and so on) are usually excellent amendments to the garden. Your first step however should be to run some basic soil tests and see what amendments are needed and in what amounts. Your county extension should be able to help you run the tests and interpret the results in keeping with that types of plants you intend to grow there.
