Primulas here come back year after year, and I would like to know when I can separate them, early spring or after they bloom and lose their flowers?

I'd wait until the flowering is finished to dig up and separate the primulas - otherwise they may stop flowering all together. They're easy to separate. Dig the entire clump then carefully divide, making sure each division has both foliage and roots attached. Replant, water well, and wait for their glorious display next spring.
