I would like to know when is the best time to prune Grapes and how to go about it. We are in Southern Indiana....

Grapes are pruned in late winter to spring. There are many different ways to train grapes so to some extent your pruning will be determined by how the grapes are supported. The general idea though is to cut them back very hard, and to develop a main stem with some selected side vines. Here are some instructions with diagrams to help you plan what to do.
