I am going to start a new garden and would like to know if I can apply Round Up to kill the heavy bluestem grass and other weeds and if I need to brake the ground beefore spraying?

I am sorry for this delayed reply to your gardening question. The spring rush has brought a deluge of questions and we are working hard to catch up!

You can put Roundup on the weeds. It works best if the weeds are actively growing. Give it a week to translocate down so it can kill the weeds and then mow the area very closely and then rototill or spade up the soil to begin preparing it for gardening.
