Will my perennials grow through a new layer of sweet peet mulch?

Mulch is generally applied over bare earth and around existing plants. If you spread it over the top of your garden bed before the perennials pop up, they many not have the strength to work their way through the mulch. I'd wait until the perennials are up and growing, then spread mulch around them.
