I have a new open-bottom composter. The place I will set it up is covered with grass. Should I remove the grass so the compost can take advantage of the microbes in the soil underneath the grass?

The grass directly beneath the composter will die out as soon as you place the composter on the lawn and begin to fill it with organic matter. So the choice is yours; you can remove the sod first, or you can simply leave it in place and it will die out on its own.
