We removed a large above ground pool and want to plant grass over the sand base. We will need to add 3-4 inches of soil to level. Should we remove the sand base first?

You won't need to remove the sand but you should mix in some topsoil and/or some compost or other organic matter to help hold moisture so your grass will thrive. Sandy soils drain well - too well to support turfgrass. Amending the soil will give the grass a healthy place to root.
