We have a 6x6' area in the yard where I do used to do his business. We no longer have him. What do we do to that area to prepare it for grass in the spring?

Dog urine contains concentrated salts which can render the soil sterile after a while. I'd either dig and remove the top few inches of soil and replace it with fresh soil from elsewhere in the garden, or rototill the area and mix in some compost which will help neutralize the salts. Either way, your new turf should thrive. Best wishes with your lawn!
