We had a wisteria planted just off the back lawn, which bloomed magnificently for several years. Every now and then a shoot would come up somewhere in the lawn, but I'd just dig down and break it off from the horizontal root... no problem. Then the plant died for some unknown reason, and now its roots have gone crazy and are putting shoots up through the lawn like there's no tomorrow. I really don't want to dig up the lawn to get the roots out, so I've been painting the shoots that come up with Roundup, hoping that the roots will get the idea. Is this a feasible means of doing the job, is there a better root-killer, or a better 'indirect' way in general?

Well, I don't know of any root killer for trees/shrubs.

But, it would help to know exactly where your plant is. Do you have any photos?
