Wow, that's really cool! It makes sense to fill the tube with soil and the rest with moss. The number of semps needed would depend on how full you want it (instant gratification), or if you have lots of patience (it'll take a year or two to fill in). If you're using only semps I'd say 40-75 depending on my above "requirements".
